چکیده
I. General methods All Fmoc-protected natural amino acids were purchased from Advanced Chemtech (Louisville, KY). Fmocperfluorophenylalanine (Fmoc-Z) was purchased from Chem-Impex Int’l Inc.(Wood Dale, IL). Fmoc-Gly-Wang resin was purchased from Novabiochem (San Diego, CA). All other chemicals were purchased from Sigma-Aldrich (Milwaukee, WI). Peptide synthesis was finished on a Tribute peptide synthesizer (Protein Technologies, Tucson, AZ). H-NMR and C-NMR data were collected on a Varian Gemini 500 MHz NMR spectrometer. HR-MS data were generated by Boston College Mass-Spec facilities. The concentration of all labeled proteins was determined in Tris buffer (10 mM Tris, 100 mM Sodium Citrate, pH=8.5) by measuring their absorption at 350 nm (ε = 13,405 M cm) on a Thermo Scientific NanoDrop 2000c (Wilmington, DE). All fluorescence data were collected on a Fluorolog spectrometer (Horiba Jobin Yvon FL3-22).

An improved reversibly dimerizing mutant of the FK506-binding protein FKBP
FK506-binding protein (FKBP) is a monomer that binds to FK506, rapamycin, and related ligands. The F36M substitution, in which Phe36 in the ligand-binding pocket is changed to Met, leads to formation of antiparallel FKBP dimers, which can be dissociated into monomers by ligand binding. This FKBP(M) mutant has been employed in the mammalian secretory pathway to generate aggregates that can be di...
متن کاملAn NMR strategy for fragment-based ligand screening utilizing a paramagnetic lanthanide probe
A nuclear magnetic resonance-based ligand screening strategy utilizing a paramagnetic lanthanide probe is presented. By fixing a paramagnetic lanthanide ion to a target protein, a pseudo-contact shift (PCS) and a paramagnetic relaxation enhancement (PRE) can be observed for both the target protein and its bound ligand. Based on PRE and PCS information, the bound ligand is then screened from the...

متن کاملAn enantiomerically pure siderophore type ligand for the diastereoselective 1 : 1 complexation of lanthanide(III) ions
A facile synthesis of a highly preorganized tripodal enterobactine-type ligand 1a-H₃ consisting of a chiral C₃-symmetric macrocyclic peptide and three tridentate 2-amido-8-hydroxyquinoline coordinating units is presented. Complex formation with various metal ions (Al(³+), Ga(³+), Fe(³+), La(³+) and Eu(³+)) was investigated by spectrophotometric methods. Only in the case of La(³+) and Eu(³+) wer...
متن کاملP-31: The Alteration of SpermatogenesisHas A Correlation with Sertoli Cell Mitochondrial Abnormal Morphology in Cytotoxicity of Testicular Tissue Mediatedwith Monosodium
Background: Male infertility has many causes, including genetic infertility. The NOP2/Sun domain family, member7 (Nsun7) gene, which encodes putative methyltransferase Nsun7, has a role in sperm motility. The aim of the present study was to investigate the effect of the T26248G polymorphism on Nsun7 protein function and its role in male infertility. Materials and Methods: Semen samples were col...
